Before she dies, May tells Parker that "with great power, there must also come great responsibility". Under Strange's suggestion, Parker tries to convince an MIT administrator to reconsider the applications of both MJ and Ned but is attacked by Otto Octavius. Octavius rips Parker's nanotechnology from his Iron Spider suit, which bonds with his mechanical tentacles and allows Parker to disable them. As Norman Osborn arrives and attacks, Strange teleports Parker back to the Sanctum and locks Octavius in a cell next to Curt Connors. Strange explains that before he could contain the corrupted spell, it summoned people from other universes within the multiverse who know Spider-Man's identity. He orders Parker, MJ, and Ned to find and capture them; they locate and retrieve Max Dillon and Flint Marko.

Thomas Haden Church reprises his role as Flint Marko / Sandman, a small-time thief from Raimi's Spider-Man 3 who received an ability to transform into sand. Cliff Robertson reprises his role as Peter's uncle Ben Parker in a flashback. On April 16, 2021, it was reported that Jon Favreau would be reprising his role as Happy Hogan in the film. That same day, Alfred Molina stated that Jon Watts told him that the incarnation of Doctor Octopus that he was portraying "would pick up" after the character's final scene in the non-MCU film Spider-Man 2.
